2011 June 8 3:30 pm Today PCR with Priyanka. We based off Anthony's last success, but with lots of changes. Don't expect it to work with so many changes, but good practice. Recipe and program are below. Notable changes 1. 100 microliter volume instead of 50 2. OneTaq buffer 5x instead of 10x 3. Used 11 microliters of dNTPs instead of 8.4 4. We mixed taq into the master mix as last step, and then added to the tubes, which had their respective primers already added (5 microliters) Started the program, it will take about 5 hours and sit at 4C overnight. https://spreadsheets2.google.com/spreads... - Steve Koch
